屏東地區臍帶血中多溴聯苯醚和甲狀腺荷爾蒙的相關性之研究 屏東地區臍帶血中多溴聯苯醚和甲狀腺荷爾蒙的相關性之研究 Characteristics of polybrominated diphenyl ethers in cord blood and their relationship with thyroid hormones in Pingtung area 研究生：吳易晉 Wu, I-Chin 指導教授：趙浩然 Chao, How-Ran 【】 現今，人類使用塑膠及電子產品越來越普遍，在市售產品皆會添加溴化阻燃劑 (Brominated flame retardants；BFRs)，目的是為了防止火災的發生。目前人類受到多溴聯苯醚 (Polybrominated diphenyl ethers；PBDEs)暴露的機會高出之前許多。數個動物實驗發現PBDEs的同構物可能會影響新生兒神經系統、甲狀腺荷爾蒙及生長激素。本研究是收集2007年到2008年屏東地區20位參加者的臍帶血，並探討臍帶血中甲狀腺荷爾蒙、生長激素及PBDEs濃度和新生兒的統計參數。本研 究分析甲狀腺荷爾蒙(T3 (triiodothyronine), T4 (thyroxine), TSH (thyroid stimulating hormones)及free T4 (free thyroxin))和IGF-1 (Insulin-like growth factors-1)濃度是使用化學冷光微粒免疫分析技術 (chemiluminescence Immunoassay；CMIA)，而分析臍帶血血清中PBDEs濃度是使用高解析度氣相 層析儀/高解析度氣相質譜儀 (high resolution gas chromatographic mass spectrometry ；HRGC/HRMS)。T3、T4、TSH及FreeT4及IGF-1的濃度分別為0.403?0.112 ng/ml, 9.65?2.00 μg/ml, 6.11?3.97 μIU/ml, 1.23?0.170 ng/dl 及68.9?23.6 ng/ml，總PBDEs的濃度為13.9?13.1 ng/g lipid，其中以BDE-209 (7.45?10.3 ng/g lipid)濃度最高。本研究將T4以分為兩組，當T4 ? 10.85 μg/ml其BDE-15, -28, -47, -49, -85及 -209的濃度顯著高於T4 ? 10.85μg/ml的這組 (p <0.001~0.05)。而將IGF-1也同樣分成兩組，則發現當IGF-1 ? 65.0 ng/ml其BDE-47的濃度顯著低於IGF-1 ? 65.0 ng/ml (p=0.02)。本研究發現臍帶血中的PBDEs和甲狀腺荷爾蒙和IGF-1有呈顯著的負相關，所以PBDEs可能會干擾甲狀腺荷爾蒙和生長激素。 關鍵字：多溴聯苯醚、新生兒、臍帶血、甲狀腺荷爾蒙、類胰島生長因子(IGF-1) 【Abstract】 Currently, human using plastic and electronic products are popular. Brominated flame retardants (BFRs) are as the additives on the surface of these commercial products on the prevention of fire. The opportunity of human exposure to polybrominated diphenyl ethers (PBDEs) is more increasing in the present compared to the past. The findings of several in-vivo studies were that the congeners of PBDEs had the adverse effects on neurodevelopment, thyroid hormone, and growth hormones in the neonatals. This study was to collect 20 individual cord bloods in Pungting area between 2007 and 2008. The levels of thyroid and growth hormones and PBDEs in cord serum and infant demographic parameters were determined in this study. The levels of thyroid hormones including triiodothyronine (T3), thyroxine (T4), thyroid stimulating hormones (TSH), free thyroxin (free T4) and insulin-like growth factors-1 (IGF-1) are analyzed by chemiluminescence Immunoassay (CMIA) . The levels of PBDEs in cord serum are analyzed by a high resolution gas chromatograph with a high resolution mass spectrometry (HRGC/HRMS). The mean levels of T3, T4, TSH, Free T4 and IGF-1 are 0.403?0.112 ng/ml, 9.65?2.00 μg/ml, 6.11?3.97 μIU/ml, 1.23?0.170 ng/dl and 68.9?23.6 ng/ml, respectively. The mean level of PBDEs was 13.9?13.1 ng/g lipid and the predominant PBDEs was BDE-209 with 7.45?10.3 ng/g lipid. The levels of BDE-15, -28, -47, -49, -85 and -209 in the group of T4 ? 10.85 μg/ml were significantly lower than those in the group of T4 ? 10.85μg/ml (p<0.001~0.05). The lower BDE-47 concentrations were found in the concentrations of IGF-1 higher than 65.0 μg/ml compared fo those of IGF-1less than 65.0 μg/ml (p=0.02). The finding of the present study was that the increased PBDEs in cord blood had the significant associations with the decreased thyroid and IGF-1 hormones.
